Reducible amino acid based cationic lipids with a naphthalimide moiety as non-viral gene vehicles

Three basic amino acid–based cationic lipids bearing a fluorescent naphthalimide moiety and reducible disulfide linkage are synthesized applied as non-viral gene vehicles. Their DNA interactions investigated by agarose-gel retardant ethidium bromide replacement assays. The sizes zeta potentials of the liposome/DNA complexes measured dynamic light scattering. cytotoxicities examined using HeLa 7702 cell lines MTT glutathione-responsive release process is studied through time-dependent fluorescence Luciferase expression showed transfection efficiency liposome dramatically increased in presence 10% serum. Confocal laser scanning microscopy studies corroborated that successfully uptaken into cells. These results demonstrate promising use acids naphthalimide-containing for safe efficient delivery.
